La melina es una especie forestal con una alta calidad maderable y de importancia económica para diferentes países tropicales como Colombia. No obstante, existe poca información acerca de su genética, lo cual es pertinente en programas de mejoramiento.

Con el fin de determinar el tamaño del genoma de Gmelina arborea (valor C), se obtuvieron suspensiones nucleares a partir de hojas, las cuales se analizaron mediante citometría de flujo. Para la obtención de suspensiones acuosas de núcleos, se evaluaron cuatro buffers de extracción, de los cuales, el buffer LB01 produjo poblaciones nucleares intactas y con mayor homogeneidad. Se analizaron tres especies vegetales como posibles estándares para la determinación del valor C; utilizando tomate como estándar referente, se obtuvo un valor C para melina de 467 Mpb. Este es el primer reporte relacionado con la estimación del valor C en melina, por lo que se genera así información importante para incrementar el conocimiento relacionado con esta especie.

Melina, also known as white teak, is a species of high timber quality and economic importance for different tropical countries like Colombia. However, information about its genetics is scarce, which is relevant for breeding programs. In order to determine the genome size of Gmelina arborea (C value), nuclei suspensions were obtained from leaves, and analyzed using flow cytometry. To obtain aqueous nuclei suspensions, four extraction buffers were evaluated, LB01 buffer produced intact populations with greater homogeneity. Three species were analyzed as possible reference standards for the determination of the C value; using tomato as a reference species, a C value for melina of 467 Mpb was obtained. This is the first report related to the estimation of the C value in Gmelina arborea, generating important information to increase knowledge related to this species
